摘要
目的探讨脂质(三酯酰甘油和极低密度脂蛋白)对 HSC前胶原 Ⅰ、 ⅢmRNA表达的影响。方法链霉蛋白酶和胶原酶原位灌流, Nycodenz密度梯度离心分离大鼠 HSC,分别用三酯酰甘油(25 mg/L)和极低密度脂蛋白(25 mg/L)共孵育 10 d后,提取 HSC的 RNA,用 Northern blot杂交观察三酯酰甘油和极低密度脂蛋白对HSC前胶原Ⅰ、Ⅲ mRNA表达。结果 HSC经三酯酰甘油和极低密度脂蛋白刺激后其前胶原Ⅰ、Ⅲ mRNA表达均明显增加。结论脂质可增加前胶原Ⅰ、Ⅲ mRNA表达,与脂肪肝肝纤维化的发生有关。
Objective To study the effect of lipids on Ⅰand Ⅲ procollagen mRNA expression of hepatic stellate cells(HSC). Methods HSC were isolated and cultured from the liver of Wistar rats by in situ perfusion with pronase and collagenase and the density gradient centrifugation with Nycodenz. HSC were incubated with triglyceride (25 mg/L) and very low-density lipid (VLDL, 25 mg/L) for 10 d, respectively. Ⅰand Ⅲ procollagen mRNA expression of HSC was de- tected by Northern blot hybridization. Results After HSC was stimulated with triglyceride and VLDL, the expression of Ⅰ and Ⅲ procollagen mRNA increased obviously. Conclusion Lipids may promote the expression of Ⅰand Ⅲ procollagen mRNA, and may be associated with fatty liver and hepatic fibrogenesis.
