摘要
以纤维堆囊菌SoF5-09为出发菌株,通过超声波和紫外诱变处理,选育得到了2株具有良好遗传稳定性的突变菌株。在此基础上应用单因素试验和正交设计对一株突变菌株产埃博霉素B的发酵条件进行了系统的研究和优化,得到了最佳的试验组合为发酵温度30℃,发酵时间为7d,接种量为8%,发酵转速200r/min,采用此条件埃博霉素B的产量达33.5mg/L。
Sorangium cellulosum SOF5-09 was used as starting strain to be mutated by ultrasonic and UV treatment. Two mutant strains with good genetic stability and high epothilone B-producing were screened. Fermentation conditions of one mutant strain were optimized by single factor design and orthogonal design. The results were as follows, fermentation temperature 30℃, fermentation time 7d, inoculum 8% and shaking speed 200rpm and under these conditions, epothilone B production reached 33.5mg/L.
