摘要
为了获得能够将褐藻酸盐水解成为单糖的褐藻酸盐裂解酶,以便于在褐藻生物质能源生产过程中对褐藻的糖化处理,对筛选分离获得的产褐藻酸盐理解解酶菌株Shewanella haliotis BP-1进行产酶条件的初步优化。初步确定了产酶的条件为温度35℃,初始pH 8.0,摇床转速200r/min,接种量为1%(v/v),装液量为500mL三角瓶装200mL,时间24h。优化后的产酶培养基为1.05%(w/v)K2HPO4,0.45%(w/v)KH2PO4,0.5%(w/v)(NH4)2SO4,2%(w/v)NaCl,0.2%(w/v)MgSO4.7H2O,0.001%(w/v)FeSO4.7H2O,0.4%(w/v)海藻酸钠。优化后菌株Shewanella haliotis BP-1在发酵培养24h后酶的比活力最高为0.92U/μg,此时发酵液的酶活性为28.1U/mL,发酵48h后达到最大酶活30.4U/mL。
To obtain the alginate lyase which can hydrolyze alginate to fermentable monosaccharide for ethanol production from brown algae, the optimization of fermentation conditions for alginate lyase from Shewanella haliotis BP-I was conducted. The optimal fermentation condition was as followed: temperature 35℃, pH 8.0, rotation speed 200r/min, inoculum 1%(v/v), shaking flask volume 200mL per 500ml and fermentation time 24h. The optimal fermentation medium was as followed: K2HPO4 1.05%(w/v), KH2PO4 0.45%(w/v), (NH4)2SO4 0.5%(w/v), NaC1 2%(w/v), MgSO4 @ 7H20 0.2% (w/v), FeSO4 · 7H2O 0.001% (w/v) and sodium alginate 0.4% (w/v). Under these conditions, the highest specific activity was 0.92U/μg after fermented for 24h and the alginate lyase activity of zymotic fluid was 28.1U/mL. The highest lyase activity was 30.4U/mL after fermented for 48h.
