摘要
目的 研究促甲状腺激素释放激素 (thyrotrophin- releasing hormone,TRH)及其受体 (TRH recep-tor,TRH- R)在大鼠睾丸组织中的表达规律和在生殖发育调节中的作用。 方法 以大鼠睾丸组织总 RNA为模板 ,依据大鼠下丘脑中的前 TRH和垂体中的 TRH- R c DNA设计引物 ,进行反转录聚合酶链式反应 (reverse tran-scription- polymerase chain reaction,RT- PCR) ,对特异性片段进行序列测定。 结果 从大鼠睾丸组织中获得了前 TRH和 TRH- R的 c DNA片段 ,其核苷酸序列与大鼠下丘脑中的前 TRH和垂体中的 TRH- R c DNA序列完全一致。 结论 首次用 RT- PCR的方法从大鼠睾丸组织中获得了前 TRH和 TRH- R的 c DNA克隆。
Objective\ In order to investigate the expression regulation of thyrotrophin\|releasing hormone(TRH) and TRH receptor (TRH\|R) in rat testis, and their function in production and development. Methods\ Total RNA of rat testis was used as template. The oligonucleotide primers were designed from the sequence of rat hypothalamus prepro(pp)TRH and pituitary TRH\|R cDNA for Reverse transcription\|polymerase chain reaction (RT\|PCR). Specific fragments of ppTRH and TRH\|R cDNA were cloned and sequenced. Results\ DNA sequence analysis indicated that cDNA sequences of ppTRH and TRH\|R from rat testis were consistent with those of hypothalamus ppTRH and pituitary TRH\|R cDNA. Conclusions\ It is the first time to acquire the cDNA cloning of ppTRH and TRH\|R from rat testis by RT\|PCR.
