摘要
目的观察水通道蛋白-3(AQP-3)在SD大鼠正常骨骼肌中的表达,为深入探讨AQP在大鼠骨骼肌肿胀状态下的表达及生理作用奠定基础。方法收集8只6月龄SD大鼠左后肢外侧股四头肌肌群中约200 mg肌肉组织,运用实时荧光定量PCR、Western blot检测AQP-3在大鼠肌肉组织中的表达。结果实时荧光定量PCR显示AQP-3 mRNA在肌肉组织中稳定表达;Western blot检测显示肌肉组织中AQP-3在36KD左右有一特异性条带。结论 AQP-3在SD大鼠正常肌肉组织中的稳定表达提示其可能参与肌肉组织内的液体平衡,对维持肌肉组织的正常生理功能、在创伤性肌肉肿胀的发病机制中可能有重要作用。
Objective To determine the expression of aquaporin3(AQP-3) in the Adult-rats'Skeletal Muscle. for the thorough discussion the expression of AQP in rat skeletal muscle swelling state and physiological role laying the foundation. Methods About 200 mg muscle tissue (8 cases) were collected from rats'left hind limb lateral quadriceps femoris muscle group. The expression of AQP-3 was detected by real-time quantitative PCR and Western blot. Results Real-time quantitative PCR showed AQP-3 mRNA was expressed in skeletal muscle. Western blot revealed there was a specific band at the site of around 36KD. Conclusion The expression of AQP-3 in rats'keletal muscle suggest that it may be involved in the liquid balance of muscle tissue, and play an important role in maintaining muscle tissue of normal physiological function, in the pathogenesis of traumatic muscle swelling.
