摘要
目的:探讨TGF-β1、Smad2在慢性哮喘大鼠肺组织中的定位表达及其含量和mRNA变化情况,以反映其在哮喘大鼠气道重建过程的作用。方法:实验设正常组、模型组、复方麻芩汤和地塞米松组。肺组织病理切片(HE染色法);苦味酸酸性复红染色法测定支气管组织胶原纤维含量;荧光免疫组化法测定TGF-β1、Smad2蛋白家族在肺组织中的定位;ELIEA法测TGF-β1的含量;RT-PCR测定TGF-β1、Smad2蛋白家族mRNA的变化情况。结果:HE染色显示模型组有较明显出血,肺泡均有一定程度扩张,有慢性炎细胞浸润;荧光免疫组化法显示TGF-β1、Smad2蛋白家族均在支气管壁和肺泡壁上表达;苦味酸酸性复红染色法显示模型组肺组织胶原纤维较正常组含量显著增高(P<0.05),复方麻芩汤组胶原纤维含量明显下降(P<0.05);ELIEA法显示模型组较正常组TGF-β1含量明显增高(P<0.05),而复方麻芩汤组、地塞米松组较模型组相比具有明显的下降(P<0.05);RT-PCR测定显示模型组的TGF-β1、Smad2的mRNA与正常组相比表达明显增加,而地塞米松能明显降低TGF-β1、Smad2的mRNA的含量,复方麻芩汤的改善效果并不明显。结论:TGF-β1蛋白与慢性哮喘关系密切,复方麻芩汤对哮喘气道重建有良好的治疗作用,能调节TGF-β1的水平,改善气道功能。
Objective: To explore changes of TGF-β1,smad2 location expression,its content and mRNA in lung tissue of chronic asthma rats,and try to reflect its effects in airway restruction of asthma rats.Methods: The experiment was set up 4 groups: normal group,model group,Ma Qin Tang compound group,dexamethasone group.The lung tissue biopsy(HE staining);picric acid fuchsin staining method for the determination of bronchial tissue collagen content;fluorescent immunohistochemical method for the determination the position of TGF-β1,Smad2;ELIEA method to measure the level of TGF-β1;RT-PCR determination of TGF-β1,Smad2 mRNA changes.Results: HE staining showed that the model group was obviously bleeding,there was a certain degree of alveolar expanding of lung alveolar and inflammatory cell.Immunofluorescence showed TGF-β1,Smad2 protein were located in the bronchial wall or alveolar wall;Bitter Acid fuchsin staining showed that the model group was significantly higher than the normal group(P0.05),followed by the rise of a dexamethasone group,but Ma Qin Tang group collagen fibers were significantly decreased(P0.05);ELIEA method showed TGF-β1 content of the model group was significantly higher than normal group(P0.05),while Ma Qin Tang group and dexamethasone group decreased significantly than model group(P0.05);RT-PCR showed that TGF-β1,Smad2 mRNA of the model group was significantly increased compared to the normal group,but dexamethasone could significantly reduced TGF-β1,Smad2 mRNA levels.Conclusion: TGF-β1 protein had a certain relationship with chronic asthma.Ma Qin Tang compound had a certain treatment to airway reconstruction of asthma,which could regulate TGF-β1 levels,improved airway function.
