摘要
[目的]优化中华被毛孢的液体培养条件。[方法]通过18srDNA及系统进化树对中华被毛孢进行鉴定分析,然后以中华被毛孢菌丝体生物量为指标,采用单因素试验和正交试验研究菌丝体液深层发酵的培养条件。[结果]单因素试验表明,最佳的氮源为蛋白胨,其次为牛肉膏;最佳碳源为葡萄糖,其次为蔗糖和麦芽糖。正交设计试验表明,菌株的液体发酵的最佳配方:浓度1%葡萄糖+浓度0.5%蛋白胨+浓度0.1%酵母粉+浓度0.01%维生素B1+浓度0.05%KH2PO4+浓度0.05%MgSO4;在该浓度培养下,菌丝体生物量可达到20.8 g/L。[结论]该方法优化了中华被毛孢的液体培养条件,为中华被毛孢的的开发利用提供依据。
[ Objective ] To optimize liquid culture conditions for Hirsutella sinenis. [ Method ] H. sinenis was identified with its molecular phy- logenetic analysis of 18S rDNA sequences. With mycelium biomass as the index, single factor test and orthogonal test were carried out for stud- ying culture conditions of mycelium deeply fermentation. [ Result] The single factor test showed that the optimum nitrogen source is peptone, followed by the beef paste ; the optimum carbon source is glucose, followed by sucrose and maltose. The orthogonal experiment showed that the optimum formula of liquid fermentation is: 1% glucose + 0.5% peptone +0.1% yeast powder + 0.01% vitamin Bt + 0.05% KH2PQ + 0.05 % MgSO4; under the above conditions, the biomass was up to 20.8 g/L. [ Conclusion ] The method optimized the culture conditions of Hirsutella sinenzis, which provides basis for the development and utilization of H. sinenis.
