摘要
目的 建立高低不同转移特性的人骨肉瘤克隆细胞株 .方法 通过克隆技术和细胞电泳 ,从我们建立的骨肉瘤细胞系 L TH- OS(简称 S)中获得两株细胞 S1 和 S2 ,测定两种细胞的增殖率 ,利用软琼脂克隆形成率和裸鼠体内成瘤实验 ,比较细胞的转移特性 .结果 S1 的细胞电泳率高达 1.0 ,S2 最低为 0 .70 ,S1 的群体倍增时间为 30 .8h,S2 为 45 .0 h.克隆形成率分别为 5 .2 %和 2 .1% ,裸鼠右后肢皮下接种 S1 和 S2 发现S1 成瘤快 ,且瘤体大 ,原位接种时 S1 出现广泛的肺转移 .结论 S1 和 S2 是从
AIM To establish two human osteosarcoma cell clone lines with high and low metastatic capacities. METHODS Two cell clones derived from human osteosarcoma cell LTH OS(S)were established in our laboratory through clone and electrophoretic technique.Cloning and xentransplantation in nude mice were employed to analyze the character of S 1 and S 2. RESULT Electrophoretic mobility of S 1 and S 2 were 1.00 and 0.70 respectively. The population doubling time was 30 8 h and 45.0h respectively. The clogenesis rates were 5.2%and 2.1% respectively. After subcutaneous transplanta tion of S 1 and S 2,S 1 showed a faster growth speed and bigger volume of the tumor than S 2. Extensive lung metastasis focus can be determined by orthotopically transplanting S 1. CONCLUSION S 1 is a high metastatic osteosarcoma cell line from LTH OS.
