摘要
目的 从犬四联弱毒样品中分离获得一株细小病毒(CPV)弱毒株 ,用于疫苗制备 .方法 根据犬副流感的血球吸附特性、细胞培养特性以及异种病毒抗血清阻断法分离病毒 .该病毒简称为 CPV- XN1 株 .并对犬细小病毒疫苗的免疫性和动物安全性进行了研究 .结果 病毒分离后其血凝效价(HA)值为 1∶ 10 2 4~ 1∶ 40 96或 10 7.0 TCID5 0 ;在电镜下可见较典型的犬细小病毒形态结构特征 ;免疫电镜下见到抗体分子已被吸附在病毒颗粒表面 ;病毒接种到健康幼犬后 15 d内未发现仔犬死亡 ;实验组犬均未出现临床症状 ;疫苗保存 6mo,其效价只降低一个滴度 .
AIM To produce a polyvalent vaccine by isolating a strain of canine parvovirus (CPV) attenuated strain from quadrivalent attenuated sample. METHODS Using erythrocyte absorption experiment, cell culture and hererovirus anti serum blocking assaying,CPV was isolated. The obtained virus was named CPV XN 1. Its immunological character and animal security were analyzed in vitro . RESULTS The HA titer of CPV XN 1 was 1∶1024~1∶4096 or TCID 50 10 -7.0 . CPV XN 1 had a typical CPV morphological character underelectron microscope, antibody molecules could be observed to absorb on the surface of virus particles. After inoculating CPV vaccine against healthy infant dogs, we found on death in 15 days. The vaccine validity only fell one titer in 6 moths. CONCLUSION This virus strain can serve as a candidate for strain of canine polyvalent attenuated vaccine.
