磷脂酶C-γ1对乳腺癌细胞运动作用的影响

摘要 目的研究磷脂酶C-γ1(phospholipase C-γ1,PLC-γ1)对乳腺癌细胞生长、黏附和迁移等生物学行为的影响,揭示乳腺癌发生的新分子机制。方法在乳腺癌MDA-MB-231细胞中加入PLC-γ1抑制剂,观察细胞划痕、趋化和黏附运动能力变化,应用Western blot技术检测相应的乳腺癌信号转导通路中整合素β1分子磷酸化的表达。结果 PLC-γ1被抑制后乳腺癌细胞的迁移和黏附运动能力下降,差异有统计学意义(P<0.05);整合素β1的磷酸化表达水平降低。结论 PLC-γ1参与了乳腺癌细胞迁移和黏附运动,可作为乳腺癌分子诊断和靶向治疗的靶点。
