摘要
研究探讨了长心卡帕藻外植体在PES、PESI、f/2三种液体培养基中的培养效果。在PES培养基中分别添加浓度为2mg/L的IAA、GA3、不同浓度梯度(0.5、2、5mg/L)的6-BA及0.5%椰子水,研究上述生长调节物质对外植体不定芽发生的影响。结果表明,培养后40 d,外植体在PES中生长最佳,死亡率为33.33%,出芽率为60%,延长培养3个月后相对生长率达到2.86%。观察发现外植体不定芽的发生具有明显的极性,3种植物激素中6-BA 2mg/L对外植体不定芽的发生有促进作用,该处理的出芽率和平均出芽数均最高,分别为90%和2.27个,IAA和GA3无明显效果。椰子水的加入则改变了培养基的物理性质,不利于外植体的生长和发芽。
The effects of PES,PESI and f/2 liquid cuhure medium were tested on tissue culture of Kappaphycus alvarezii, regeneration from explants and form shoots in the four plant growth regulators : IAA (2mg/L),GA3 (2rag/L), 6-BA (0.5,2,5mg/L)and coconut water (0.5%). After 40 days, explants cultured in PES had the lowest death rate (33.33%) and highest shooting rate (60%); Extending the culture after 3 months, the relative growth rate was 2.86%. Explant cultured in PES with 6-BA (2mg/L) showed highest shooting rate(90%) and average shooting number(2.27) but had not improvement in IAA and GA3. Addition of coconut water changed the medium' s physical form which inhibited the regeneration of explants.
