摘要
目的 :探讨白细胞介素 - 6 (IL - 6 )对人脐静脉内皮细胞 (HUVEC)合成前列环素 (PGI2 )的影响以及钙拮抗剂、糖皮质激素对 IL - 6的作用的影响。方法 :采用培养的原代人脐静脉内皮细胞为实验对象 ,用放免法测定细胞培养液中的 6 - keto PGF1α。结果 :IL - 6组内皮细胞合成的 6 - keto PGF1α的量较对照组显著减少 ,对照组为 (6 2 0 .4±34 .1) pg/ ml,而 IL - 6组 (5 0 u/ m l、10 0 u/ ml)分别为 (5 0 3.2± 2 3.6 )、(470 .3± 2 3.6 ) pg/ m l(P <0 .0 5 ) ;钙拮抗剂可以拮抗 IL - 6的上述效应与 IL - 6组比较 ,P <0 .0 5 ,而地塞米松无此作用。结论 :IL - 6能抑制内皮细胞合成分泌PGI2 。
Objective:Our purpose was to determine the effect of interleukin 6 (IL 6) on the synthesis of PGI 2 in endothelial cells and the influence of calcium antagonist Nicardipine on the effect of IL 6.Methods: We put IL 6, monoclonal antibody of IL 6, and Nicardipine on cultured human umbilical veins endothelial cells(HUVEC) and then measured the concentration of PGI 2 in cultural fluid by radioimmunoassay.Results: The concentration of PGI 2 in IL 6 (50u/ml,100 u/ml) group was significantly lower than that of control [(503.2±23.6) pg/ml, (470.3 ±23.6) pg/ml, and (620.4±34.1) pg/ml ,P<0.05)]. That of IL 6 plus antibody or Nicardipine group were recovered to (470.3±20.3)pg/ml, (591.7±27.9)pg/ml, and (548.9±20.9) pg/ml respectively (P< 0.05 ) .Conclusion:IL 6 inhibited PGI 2 synthesis in HUVEC, monoclonal antibody of IL 6 and Nicardipine blocked this effect of IL 6 and calcium might be the second messenger in this process .
