摘要
目的 观察上皮细胞钙粘蛋白 (E cad)表达与膀胱移行细胞癌 (TCC)分级、分期等生物学行为的关系。方法 采用恒冷切片免疫组织化学SP法 ,对 2 8例TCC和 10例正常膀胱粘膜新鲜冰冻标本进行E cad表达的检测。结果 对照组 10例正常膀胱粘膜E cad均正常表达 ,实验组 2 8例TCC ,仅 50 % (14 2 8)正常表达 ,二组差异显著 (P <0 0 1)。TCC不同分级、分期间E cad表达有显著性差异 (P <0 0 5) ,G3 比G1+G2 表达低 ,浸润性TCC比浅表性TCC表达低。初发与复发、单发与多发TCC之间比较 ,E cad表达均未见显著性差异 (P >0 0 5)。本组未见E cad表达完全缺失的TCC。结论 正常膀胱粘膜组织E cad保留表达 ,TCCE cad表达降低 ,E cad表达与TCC分级、分期呈负相关 ;E cad表达与TCC的数目 。
Objective To investigate the relationship between E cadherin(E cad)expression and histopathological grade,tumor stage and the other biological behavior in transitional cell carcinoma of urinary bladder.Methods Immunohistochemical streptavidin peroxidase method(SP) was employed using a mouse monoclonal antibody for human E cad(HECD 1)to stain serial snap frozen sections of 10 normal bladder and 28 TCC specimens.Results E cad expression was normal in 50%(14/28) of TCC s versus in 100%(10/10)of normal bladder specimens( P <0 01).Abnormal expression of E cad was significantly correlated with grade and stage of TCC,26 3%(5/19) of grade Ⅰ and grade Ⅱ TCC sshowed abnormal E cad expression compared with 100%(9/9) of grade Ⅲ TCC s( P <0 001),abnormal expression was found in 87 5%(7/8) of invasive TCC s compared with 35%(7/20) of superficial TCC( P <0 05).The differences were not statistically significant between initial tumor with recurrent tumor and solitary tumor with multifocal tumor ( P >0 05).All of 14 TCC swith abnormal E cad expression displayed heterogeneous staining, completely negative TCC had not been observed.Conclusion E cad expression in normal bladder transitional epithelia was preserved whereas TCC showed a reduction of E cad expression.Decreased E cad expression was associated with both increased grade and stage of TCC.E cad expression didnt correlate with the number of TCC,no difference was found between initial TCC with recurrent TCC.
