凋亡基因ras突变和rasP21蛋白表达与尿路上皮肿瘤预后关系

Ras Gene Mutation and Ras P21 Protein Overexpression and DNA Abnormality in Urothelial Tumors Correlated with Its Prognosis

摘要 目的 研究凋亡基因ras突变和rasP2 1蛋白表达及DNA倍体异常与尿路上皮肿瘤预后关系。方法 应用聚合酶链式反应加单链多态性分析 (PCR SSCP)及流式细胞术和细胞免疫荧光技术检测 52例尿路上皮肿瘤K ras基因突变及rasP2 1蛋白表达和DNA含量变化。K ras基因突变5例 (9 6 % ) ,rasP2 1蛋白阳性表达 36例 (6 9 2 % ) ;异倍体 2 2例 (4 2 3% ) ,二倍体 30例 (57 7% ) ;DNA倍体异常和rasP2 1蛋白表达随肿瘤病理分级和分期的上升而增加 ,与肿瘤预后呈正相关 (P <0 0 1) ;异倍体肿瘤其rasP2 1蛋白表达率明显高于二倍体肿瘤 (P <0 0 1)。结论 DNA倍体异常和rasP2 Objective To study K ras gene mutation and RasP21 protein expression and DNA ploidy abnormality and find their correlation with prognosis in 52 cases of urothelial tumors.Methods K ras gene mutation was detected by polymerse chain reaction and single stranded comformation polymorphism and RasP21 and DNA content using flow cytomytry and celluar immunofluosecence technique from 52 cases of urothelial tumors.Results K ras mutation was oberverd in 9 6% and the positive expression rate of rasP21 protein in 69 2% 42 3% aneuploidy and 57 7% diploidy tumors were detected.DNA ploidy abnormality and the positive expression of rasP21 protein increased well with rise of tumor grade and stage,but had a worse prognosis( P <0 01).Conclusion DNA ploidy abnormality and rasP21 protein overexpression have been shown to be siginificant predictors of the prognosis of urothelial tumor.
