摘要 探索用化学免疫方法防治梭曼中毒。方法 :提出“潜过渡态”半抗原设计策略 ,并据此设计、合成了新型半抗原和人工抗原。结果 :该新型半抗原可成功诱导具有类酶活性的催化梭曼水解的抗体酶 ;还发现了一种新型的磷酰酯交换反应。结论 :实验证明了该半抗原设计策略的有效性。该半抗原设计策略还可能应用于能诱生催化磷 (膦 )酸二酯水解的抗体酶的半抗原设计 ,以及应用于有类似酯酶、酰胺酶、肽酶或核苷酸活性的抗体酶的研究。 Objective: To explore novel strategy for soman antidote by chemical immunization. Methods:A new hapten design strategy termed “pro transition state”hapten strategy was proposed and a novel hapten and relative synthesized antigens were designed and synthesized. Results: The synthesized antigen induced catalytic antibodies with high efficiency in catalyzing hydrolysis of nerve agent soman and a novel phospho transesterification reaction was discovered. Conclusions: The experiments demonstrated that very potent effects could be achieved through the pro transition state hapten strategy. It may be an alternative strategy for hapten design of antibody catalysis involved in the hydrolysis of nucleotides, esters, carbamates, amides and peptides. [
