摘要
目的 研究体外培养的人成纤维细胞衰老时可诱发凋亡的能力。 方法 用丁酸钠和去血清两种方法诱导人衰老和年轻成纤维细胞 ,以细胞生存曲线、细胞形态和超微结构、DNA断裂电泳图谱、流式细胞仪分析凋亡峰等手段检测凋亡的发生情况。 结果 5 0 mm ol/ L的丁酸钠诱导年轻细胞 48h即发生凋亡 ,而衰老细胞需 96 h;去血清诱导年轻细胞 15 d左右发生凋亡 ,而衰老细胞此时尚未出现凋亡现象。 结论 人衰老成纤维细胞可诱发凋亡的能力明显低于年轻细胞。
Objective To study the ability of inducing apoptosis in senescent human fibroblasts cultured in vitro. Methods Senescent and young human fibroblasts were induced by sodium butyrate (NaBu) or serum deprivation. Apoptosis was observed from analysis of cell survival curve, cell morphological and ultrastructural changes, DNA fragmentation electrophotogram and flow cytometry. Results 50 mmol/L NaBu induced apoptosis in young cells after 48 hours; while in senescent cells it happened after 96 hours. Serum deprivation induced apoptosis in young cells after about 15 days; while in senescent cell, no apoptotic phenomena showed at this time. Conclusions The ability of inducing apoptosis in senescent human fibroblasts is apparently lower than that in young ones.
