摘要
目的研究家族性腺瘤性息肉病患者血清蛋白质的变化,建立灵敏度和特异度高的家族性腺瘤性息肉病诊断模型。方法利用SELDI-TOF-MS技术对18例家族性腺瘤性息肉病患者术前、术后血清蛋白质分别进行检测,IMAC30芯片结合蛋白质后,用蛋白芯片仪读取数据,分析得到区分两组的树状分类规则,并构建诊断模型。结果 M8514.89蛋白质组成的模型可将两组准确分组,灵敏度和特异度分别为88.89%(16/18)和100%(18/18)。结论 SELDI-TOF MS技术可将家族性腺瘤性息肉病术前、术后正确分组,可应用于家族性腺瘤性息肉病的诊断、鉴别、随访和手术时机的选择。
Objective To research the serum protein of FAP patients, and establish the diagnostic model of FAP with high sensitivity and specificity in application. Methods Samples of sera with 18 preoperative and postoperative FAP patients were detected by surface enhanced laser desorption/ionization time of flight mass spectrometry SELDI-TOF MS, After the surface of chips IMAC30 bind the serum protein, proteinchip reader of ciphergen inc, a part of SELDI-TOF MS, would detect the signs. Then the classification rule of tree was discoverer. The model of identification of FAP would be constructed by the classification rule of tree. Results At the key M/Z values of M8514.89 protein contents of the two classes are obviously different by the software analysis. And corresponding sensitivity was 88.89% (16/18) and corresponding specificity was 100% (18/18). Conclusion THE SELDI-TOF MS could be used in the diagnosis, identification, follow-up and operative juncture of FAP.
