海参皂苷对小鼠造血调控因子的影响

INFLUENCE OF SAPONIN OF PEARSONOTHURIA GRAEFFEI ON HEMATOPOIETIC MODULATORS IN MICE

摘要 目的探讨革皮氏海参皂苷(saponin of Pearsonothuria graeffei,PGS)对骨髓抑制模型小鼠造血调控因子的影响。方法雄性ICR小鼠随机分为正常对照组、模型对照组和PGS低、高剂量组。剂量组小鼠灌胃不同浓度PGS(10和30 mg/kg),正常和模型对照组灌胃生理盐水,连续16d。除正常对照组外,其余小鼠均于首次PGS后第7d,连续3d腹腔注射环磷酰胺(100 mg/kg),建立骨髓抑制小鼠模型。于末次给药后,分别制备小鼠肺条件培养液(LCM)、脾条件培养液(SCM)和腹腔巨噬细胞条件培养液(PMΦCM),采用MTT法检测其对正常大鼠骨髓细胞增殖的影响;采用单层半固体培养法检测对粒单系祖细胞(CFU-GM)、红系祖细胞(CFU-E)和巨核系祖细胞(CFU-Meg)集落形成的影响;采用RT-PCR法检测粒/单核细胞集落刺激因子(GM-CSF)、促红细胞生成素受体(EPOR)和血小板生成素(TPO)mRNA的表达。结果经PGS体内诱导所制备的LCM、SCM、PMΦCM条件培养液能显著促进正常小鼠骨髓细胞增殖,增加CFU-GM、CFU-E和CFU-Meg祖细胞的集落形成,上调骨髓细胞GM-CSF、EPOR和TPO mRNA的表达量。结论 PGS通过诱导造血细胞因子的分泌促进骨髓粒单系、红系与巨核系造血的恢复。[营养学报,2012,34(6):599-604] Objective To evaluate the influence of saponin of Pearsonothuria graeffei(PGS) on hematopoietic modulators in myelosuppression mice.Method Male ICR mice were randomly divided into four groups: normal control group,model control group,low and high dose PGS groups.PGS was orally administered with 10 and 30 mg/kg.bw every day for 16 d.The mice in normal and model control groups were administered with 0.9% NaCl solution as placebo.On the 7th day,all mice except those in the normal group were injected intraperitoneally with 100 mg/kg cyclophosphamide once a day for 3 d to induce myelosuppressed animal model.The lung conditioned medium(LCM),spleen cell conditioned medium(SCM) and peritoneal cavity macrophage conditioned medium(PMΦCM) treated with Pearsonothuria graeffei were prepared to observe the effects on proliferation of bone marrow cells and formation of hematopoietic progenitor cells colonys including granulocyte-monocyte colony forming units(CFU-GM),colony-forming units-erythroid(CFU-E) and colony forming unit-megakaryocyte(CFU-Meg) by MTT.The mRNA expressions of granulocyre-macrophage colony-stimulating factor(GM-CSF),erythropoietin receptor(EPOR) and thrombopoietin(TPO) were determined by semi-quantity RT-PCR.Results PGS could significantly promote the proliferation of bone marrow cells,the colony formation of CFU-GM,CFU-E,CFU-Meg in normal mice,but each colony showed various features.PGS could increase mRNA expression abundance of GM-CSF,EPOR and TPO in bone marrow cells.Conclusion Saponin can promote effectively the hematopoietic function by up-regulating level of hemopoietic growth factors in myelosuppressed mice.[ACTA NUTRIMENTA SINICA,2012,34(6):599604]
