摘要
嗜盐隐杆藻 ( Aphanothece halophytica)培养液经离心、浓缩、透析、有机溶剂沉淀得胞外多糖 ( Exopolysaccharides,EPS)粗品 ,经 DEAE-纤维素二次柱层析纯化得EPS精品。葡聚糖 G- 2 0 0凝胶过滤表明其为单一组份。对其进行理化测试并对各组分进行定量分析 ,多糖、己糖醛酸、硫酸根含量分别为 4 0 .96% ,2 3.2 7%和 34 .4 6%。元素分析仪测得 C、H、N、S含量分别为 10 .68% ,3.4 2 % ,0 %和 7.32 % ,EPS特性粘度为 1.0 65( 10 0 ml/ g) ,旋光率为 - 2 6.316°。
The crude exopolysaccharide was isolated from the cell culture of Aphanothece halophytica after the medium being centrifugated,evaporated,dialyzed and precipitated with ethanol.The pure EPS was obtained by two times of purification on DEAE- cellulose columns.It was identified by Sephadex G- 2 0 0 column chromatography to be homogenous. Chemical analysis showed thatthe polymer contained carbohydrate,uronic acid and sulphated groups,which account for40 .96 % ,2 3.2 7% and 34.46 % of the pure EPS weight.The element analysis showed that the contentof C、H、N and Sis respectively of 1 0 .6 8% ,3.42 % , 0 % and7.32 % .The nature viscosity of EPS was1 .0 6 5 ( 1 0 0 ml/g) and the specific rotation was - 2 6 .31 6°.
