摘要
目的:探讨存活素(Survivin)和环氧合酶-2(COX-2)在视网膜新生血管中的表达。方法:以高浓度氧诱导C57BL/6J小鼠建立视网膜新生血管模型。取对照组和给氧组幼鼠眼球作荧光素血管灌注、病理切片及免疫组织化学检测,分别观察视网膜血管的改变、视网膜新生血管内皮细胞数目及Survivin和COX-2蛋白的表达。结果:给氧组视网膜可见大量新生血管形成,新生血管内皮细胞核数为23.38±1.07个,与对照组相比差异有显著统计学意义(P<0.01)。正常对照组:视网膜组织各层中未见Survivin和COX-2蛋白表达(2.56±0.46,2.37±0.80)。给氧组:Survivin蛋白表达在神经节细胞层和突破视网膜内界膜的新生血管(3.34±0.40),COX-2蛋白表达在内核层、神经节细胞层和突破视网膜内界膜的新生血管(3.89±0.56)。两者表达呈显著正相关(r=0.394,P<0.01)。结论:在视网膜新生血管形成中存在Survivin和COX-2的高表达,且两者表达密切相关。
AIM:To explore expression and relationship of COX-2 and Survivin protein in the retinal neovascularization.METHODS:Mouse model of hyperoxia-induced ischemic retinopathy were established.A novel flucorescein-dextran perfusion method was developed to assess the vascular pattern.Histologic methods were used to count blood vessel profiles in the inner retina with HE staining.Survivin and COX-2 expressions in the retina were determined by immunohistochemical method.RESULTS:Lots of neovascularization were seen in hyperoxia group.The number of nuclei of proliferative retinal vessels increased significantly as compared with normal control group(P0.01).The expression of Survivin was noted in the ganglion cells and the neovascularization breaking through the inner retinal.The expression of COX-2 was noted in the inner nuclear layer and ganglion cells and retinal blood vessels.The expressions of them have positive correlation(r=0.394,P0.01).CONCLUSION:There are high expressions of Survivin and COX-2 protein in the retinal neovascularization and they have correlation.
