慢性高原红细胞增多症患者中性粒细胞吞噬能力及呼吸爆发功能 被引量:4

摘要 目的通过与高原地区健康人相比较,探讨青海慢性高原红细胞增多症(HAPC)患者外周血多形核中性粒细胞(PMN)的吞噬能力及呼吸爆发功能。方法①用全自动血细胞分析仪检测HAPC患者红细胞数、血红蛋白及白细胞分类数等;②用流式细胞仪检测中性粒细胞对FITC标记细菌的吞噬能力;③用双乙酰基二氯荧光素(DCFH-DA)荧光探针法检测中性粒细胞呼吸爆发功能。结果①HAPC患者的红细胞数、血红蛋白及红细胞压积均高于健康组,但白细胞百分率、中性粒细胞百分率、淋巴细胞百分率及单核细胞百分率与健康组相比无明显差异;②机体在缺氧状态下,PMN吞噬能力及呼吸爆发能力降低,HAPC患者尤为明显。结论高原缺氧在一定程度上降低PMN的吞噬和杀伤能力。 Objective To investigate the changes of ability of phagocytosis and the function of respiratory burst of polymorphic neutrophils(PMN)in patients with High Altitude Polycythemia(HAPC)by comparison with healthy individuals living in plateau area at Qinghai province.Methods To analyze the red blood cells(RBC)count,hemoglobin(HGB)determination and the leukocyte classification count by automatic blood cell analyzer;To detect the ability of phagocytosis of neutrophils the FITC labeled bacteria by flow cytometry;Diacetyl Base Dichloroflurescein probe was used to detect the respiratory burst function of neutrophils.Results ① the RBC count,the levels of HGB and the hematocrit in patients with HAPC were higher than those in healthy control,but the white blood cell(WBC)count,neutrophil percentage(NEUT%),lymphocyte percentage(LYMPH%)and the monocyte percentage(MONO%)were no obvious difference compared with healthy control.② In the situation of hypoxia,the ability of phagocytosis and the respiratory burst function of PMN was reduced,especially in patients with HAPC.Conclusion The present study has found that the plateau hypoxia leading the ability of phagocytosis and the killing function of PMN are reduced in some degree.
