摘要
目的了解腹腔脂肪型丝氨酸蛋白酶抑制剂(Vaspin)与脂联素(Adiponectin,APN)mRNA在肥胖及糖尿病大鼠肝脏中的表达及意义。方法雄性Wistar大鼠100只,适应性饲养1周后,随机分为三组:基础对照组15只喂以基础饲料;肥胖组20只喂以高脂饲料;糖尿病组47只喂以高脂饲料,4周后一次性腹腔注射链脲佐菌素(STZ)制备糖尿病大鼠模型。三组大鼠共饲养28周,采用半定量RT-PCR法检测大鼠肝脏组织中Vaspin及APN mRNA的表达。结果肥胖与糖尿病组大鼠Vaspin mRNA表达水平高于对照组(P<0.05),APN mRNA表达水平低于对照组(P<0.05),肥胖组Vaspin和APN mRNA表达与糖尿病组相比无统计学意义(P>0.05);Vaspin与APN mRNA表达呈中度负相关(P<0.05)。结论 Vaspin与APN mRNA表达的改变可能是糖脂代谢紊乱的一种适应性改变。
Objective To study the significance of Vaspin mRNA and Adiponectin mRNA expression in obese and diabetic rat Models.Methods One hundred male Wistar rats were accommodated to feed with normal chow for a week and then divided into three groups randomly:the control group(NC group,n=15)fed with normal chow;the diet-induced obese group(DIO group,n=20)fed with high-fat diet;the diabetic group(DM group,n=47)were fed with high-fat diet for 4 weeks and then injected strptozotocin(STZ) into abdominal cavity to induce diabetic model.The control group were injected equivalent normal saline.All rats were fed for 28 weeks.Expressions of vaspin and adiponectin mRNA in liver tissue in the 3 groups were investigated by RT-PCR.Results Compared with control group,the expression level of vaspin mRNA was higher(P0.05),but adiponectin mRNA was lower in obesity group and diabetes group(P0.05).The expression level of vaspin mRNA and adiponectin mRNA was no statistical significant difference between the obese and diabetic group(P0.05).There was moderate negatively correlated between vaspin mRNA and adiponectin mRNA in the liver tissue(P0.05).Conclusion The expression of Vaspin and Adiponectin mRNA in obese and diabetic rats may be an adaptation changes for the glucolipid metabolism.
