摘要
目的建立蒲黄及蒲黄炭的HPLC指纹图谱,对其色谱峰进行指认与比较,为蒲黄炭的质控提供依据。方法采用HPLC法,Agilent TC-C18(5μm,250mm×4.6mm);流动相为乙腈-0.05%磷酸溶液梯度洗脱;体积流量0.8mL/min;检测波长254 nm;进样量20μL。应用中药色谱指纹图谱相似度评价系统对不同产地蒲黄药材及蒲黄炭的HPLC图进行评价。结果蒲黄和蒲黄炭色谱图中各色谱峰分离良好,分别得到15和18个共有峰。结论其中6个主要色谱峰经对照品确认为香蒲新苷、异鼠李素-3-O-新橙皮糖苷、槲皮素、柚皮素、山柰酚和异鼠李素,蒲黄炒炭后指纹图谱中有3个新成分生成。
AIM To establish the chromatographic fingerprint of Typhae Pollen and Typhae Pollen carbonisata by HPLC-DAD for the quality control. METHODS Experiments of chromatographic fingerprint analysis were car- ried out using an Agilent TC-Cls column (5 μm, 250 mm ×4. 6 mm) with the mobile phase consisted of acetonitrile-0.05% phosphoric acid solution in gradient elution mode, the flow rate of 0. 8 mL/min and sample volume of 20 μL each time. Chromatographic wavelength was set at 254 nm. The Similarity Evaluation System for Chromato- graphic Fingerprint of Traditional Chinese Medicine was used to evaluate the similarity of different original Typhae Pollen and Typhae Pollen carbonisata. RESULTS Fifteen and eighteen common peaks were obtained in the fingerprints of Typhae Pollen and Typhae Pollen carbonisata, respectively. CONCLUSION Among the obtained common peaks, six peaks are identified as typhaneoside, isorhamnetin-3-O-neohesperidoside, naringenin, querce- tin, kaempferol and isorhamnetin by comparing with their reference substances. There are significant differences among the contents and constituents after being processed, especially three new peaks appeared.
