摘要
目的:研究左归饮煎剂对围绝经期大鼠卵巢细胞凋亡指数(AI)、天冬氨酸特异性半胱氨酸蛋白酶(Cysteinyl aspartate-specific protease,Caspase)-3和凋亡相关因子(Fas)表达的影响。方法:采用围绝经期大鼠模型,分别灌胃高、中、低剂量(31.00、20.67、13.78g/kg)左归饮煎剂,每周1次,连续8周。用TdT介导原位缺口末端标记法观察卵巢细胞AI;用RT-PCR和免疫组化法检测卵巢Caspase-3、FasmRNA及蛋白的表达情况。结果:高、中、低剂量左归饮煎剂可显著降低围绝经期大鼠卵巢细胞AI(P<0.01或P<0.05),显著降低围绝经期大鼠卵巢Caspase-3与Fas的表达(P<0.01)。结论:左归饮煎剂可通过抑制围绝经期大鼠卵巢细胞过度凋亡来改善卵巢功能,这种抑制作用可通过下调围绝经期卵巢Fas和Caspase-3的表达实现。
OBJECTIVE:To investigate the effects of Zuoguiyin decoction on ovarian apoptosis index and the expression of ovarian cysteinyl aspartate-specific protease(Caspase-3) and Fas in perimenopausal rats.METHODS:Perimenopausal rat model were treated by intragastric administration(ig)with low(13.78 g/kg),middle(20.67 g/kg)and high(31.00 g/kg)dose of Zuoguiyin decoction once a week for 8 weeks.TUNEL was used to evaluate ovarian apoptosis index.The mRNA and protein expression of Caspase-3 and Fas were detected by RT-PCR and immunohistochemistry,respectively.RESULTS:The apoptosis index of ovary cells in perimenopausal rats decreased obviously by high-dose,medium-dose and low-dose of Zuoguiyin(P〈0.01 or P〈0.05);the expression of Caspase-3 and Fas decreased obviously(P〈0.01).CONCLUSION:Zuoguiyin decoction could improve ovarian function in perimenopausal rats by inhibiting excessive ovarian apoptosis.And it could play this role by down-regulating the expression of ovarian Caspase-3 and Fas.
