摘要
目的:探讨DJ-1基因mRNA及其蛋白在子宫内膜癌、癌旁组织及正常子宫内膜组织中的表达规律及临床意义。方法:应用逆转录聚合酶链反应(RT-PCR)、蛋白印迹(Western blot)及免疫组织化学Elivision法检测100例子宫内膜癌、癌旁组织及30例正常子宫内膜组织中DJ-1基因mRNA及其蛋白的表达情况,分析DJ-1基因在子宫内膜癌组织中的表达与肿瘤的病理类型、分化程度、浸润深度、淋巴结转移、期别及患者血CA125水平等临床病理特征之间的关系。结果:子宫内膜癌组织中的DJ-1 mRNA及DJ-1蛋白的表达均明显高于癌旁组织及正常子宫内膜组织,癌旁组织及正常子宫内膜组织间表达无显著差异。DJ-1 mRNA的表达与子宫内膜癌的病理类型、期别及血CA125水平无明显相关性;中低分化的子宫内膜癌组织中DJ-1 mRNA的表达高于高分化者,深肌层浸润的子宫内膜癌组织中DJ-1 mRNA的表达高于局限于黏膜层及浅肌层浸润者,DJ-1 mRNA在有淋巴结转移的患者中的表达高于无淋巴结转移者。结论:DJ-1 mRNA及蛋白在子宫内膜癌组织中高表达,且可能参与了子宫内膜癌的发展、浸润和转移等环节的调控。
Objective: To investigate the expression regular pattern and clinical significance of DJ - 1 gene mRNA and its protein in endometrial carcinomas, paraneoplastic tissues and normal endometfial tissues. Methods: Reverse transcription polymerase chain reaction ( RT - PCR), Western blot and immunohistochemical Elivision methods were used to detect the expressions of DJ- 1 gepe mRNA and pro- tein in tumor tissue, para -tumor tissue of 100 cases of endometrial carcinomas and 30 cases of normal endometrial tissue, then to analyze the relationship between DJ - 1 gene and clinical pathological characteristics of endometrial carcinoma specimens, such .as:. tumor histological type, differentiation, depth of invasion, lymph node metastasis, stage and serum CA125 Ievel. Results: DJ - 1 mRNA and,it's protein ex- pression of endometrial carcinomas were significantly higher than para - tumor tissues'and normal endometrial tissues, but the expressions be- tween paraneoplastic tissues and normal endometrial tissues had no significant difference. There were no significantly relationships between DJ - 1 mRNA expression and histological type of endometrial cancer, stage, blood CA125 levels. DJ - lmRNA expressions of poorly differ- entiated endometrial carcinoma was higher than that of Well - differentiated endometrial carcinoma. DJ - 1 mRNA expressions of deep myo- metrial invasion in endometrial carcinoma was higher than that of limiting to the mucosa and superficial myometrial invasion endometrial carci- noma, DJ - 1 mRNA expressions in patients with lymph node metastasis was higher than that without lymph node metastasis. Conclusion: DJ - 1 mRNA and itg protein in endometrial carcinomas are high expressions, and may be involved in the regulations of the development , in- vasion and metastasis of endometrial cancers.
