摘要
目的研究枸杞子中2-O-β-D-葡萄糖基-L-抗坏血酸(AA-2βG)对晶状体氧化损伤的保护作用。方法体外培养正常兔晶状体,采用H2O2氧化损伤法诱导建立晶状体混浊模型,分别于24,48,72 h培养结束后观察各组晶状体(空白对照组、H2O2氧化损伤组、维生素C组、AA-2βG组)的混浊情况,检测各组晶状体组织的总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)活性、谷胱甘肽(GSH)及丙二醛(MDA)的含量变化。结果随时间延长,空白对照组晶状体混浊不明显,H2O2组混浊程度逐渐加重,Vc组和AA-2βG组晶状体混浊明显延迟;和H2O2组相比,Vc组和AA-2βG组SOD、GSH及T-AOC水平都有提高,MDA的含量降低。结论 AA-2βG可以延缓H2 O2诱导的兔晶状体混浊,对晶状体的氧化损伤具有保护作用。
Objective To investigate the effect of AA-2βG in protecting lens against oxidative damage.Methods Cultured normal rabbits lenses in vitro were randomly divided into 4 groups:control group(exclusive of H2O2),H2O2 group(oxidative damage group),H2O2 added Vc group and H2O2 added AA-2βG group.After incubation for 24h,48h and 72h,the transparency of all lenses was observed respectively;and the levels of superoxide dismutase(SOD),glutathione(GSH),total antioxidation ability(T-AOC),and malondialdehyde(MDA) were determined by spectrophotometer.Results Compared with the control group,the opacities of lens in H2O2 group was observed in earlier incubation time;the opacities of lens in the Vc group and AA-2βG group were apparently slighter than in the H2O2 group.Furthermore,compared with those in H2O2 group,the level of T-AOC,GSH and the activity of SOD in Vc group and AA-2βG group were increased statistically except the level of MDA significantly decreased.Conclusion AA-2βG can delay the lens opacification and maintain them transparency,suggesting that AA-2βG may protect lens against oxidative damage.
