摘要
目的优化并建立以高效液相色谱法测定补骨脂中补骨脂素含量的方法。方法采用超声法从补骨脂中提取得到补骨脂素,经薄层层析法、紫外分光光度法等方法进行定性分析并采用高效液相色谱法(HPLC)测定补骨脂素的含量。色谱柱为YWG C18(250 mm×4.6 mm,10μm),流动相为甲醇,检测波长246nm,柱温为30℃,流速为1 ml.min-1。结果测定补骨脂素线性范围为0.03~0.36μg.ml-1。相关系数r=0.999 1,平均加样回收率为99.1%,RSD为1.9%(n=9)。结论该方法简便、准确、可靠,可作为补骨脂中补骨脂素含量的测定方法,为补骨脂的进一步开发利用和有效性研究提供理论依据。
Objective To optimize and develop a method to determine the content of Psoralen in Psoralea corylifolia L.by HPLC.Methods The extraction of Psoralen was performed using ultrasonic extraction technology,the production was identified by TLC and UV-spectrophotometry and was determined by HPLC.Using YWG C18(250 mm×4.6 mm,10 μm)column,and methanol as the mobile phase,the flow rate was 1.0 ml·min-1,the detecting wavelength was at 246 nm,and the column temperature was 30℃.Results The linear range of Psoralen was 0.03~0.36 μg·ml-1(r=0.999 1) and the recovery rate was 99.1%,RSD 1.9%(n=9).Conclusion The method is simple and rapid to determine the content of Psoralen in Psoralea corylifolia L.and it provides scientific basis for the safety and effectiveness of Psoralea corylifolia L.in its clinical use.
