摘要
目的探讨姜黄素对人肾透明细胞癌细胞786-O体外增殖及其诱导细胞凋亡的影响。方法 MTT法观察对癌细胞生长的影响,DAPI凋亡染色检测姜黄素对细胞凋亡的诱导作用,Western Blot检测抑凋亡蛋白Bcl-2的表达。结果MTT结果表明姜黄素对细胞786-O具有较好抑制作用,25μmol/L、50μmol/L、100μmol/L浓度的姜黄素处理48h对细胞增殖抑制率分别为(45.90±1.78)%、(79.76±1.49)%、(91.11±1.38)%,并呈现一定剂量依赖关系;凋亡染色显示细胞在作用48h后,有典型的凋亡形态出现,且随着浓度的增大作用更为明显;Western Blot显示姜黄素可下调Bcl-2蛋白表达,50μmol/L和100μmol/L浓度作用较为明显。结论姜黄素可抑制人肾透明细胞癌细胞786-O的生长,并能诱导其凋亡,其作用机制可能与下调Bcl-2蛋白表达有关。
Objective To study the effect of curcumin on proliferation,inhibition,and apoptosis in human renal clear cell carcinoma 786-O cells.Methods The inhibition of cell proliferation was measured by MTT assay.The effect of inducing apoptosis was inspected by DAPI fluorescent staining.The expression of anti-apoptosis protein Bcl-2 was detected by Western Blot.Results MTT results showed that the cells growth was inhibited with the increasing of the concentration of curcumin after 48h,and the inhibition rates of cells treated by the concentration with 25、 50、100 μmol/L were(45.90±1.78)%,(79.76±1.49)%,(91.11±1.38)%,respectively.Meanwhile it showed a dose-dependent manner.Most of cells presented the typical morphological changes of apoptosis under the fluorescent microscope.The effect showed more obvious with the increasing of the concentration.The expression level of Bcl-2 was down-regulated by curcumin,significantly treated by concentration with 50μmol/L and 100μmol/L.Conclusion Curcumin can inhibit proliferation and induce apoptosis of humanmel renal clear cell carcinoma 786-O cells in vitro.The mechanism may be related to down-regulation of the expression of Bcl-2 protein.
