金钗石斛多糖的组织化学定位研究

Histochemistry Localization for Polysaccharide in Dendrobium nobile Lindl

目的优化金钗石斛多糖组织化学定位方法,明确金钗石斛多糖的分布和积累特征。方法采用徒手切片和冰冻切片方法制作金钗石斛根、茎和叶片横切片,利用组织化学定位方法对金钗石斛多糖进行定位分析。结果徒手切片法比冰冻切片法更适用于金钗石斛多糖的组织化学定位研究,金钗石斛多糖主要分布在根的皮层薄壁细胞、茎的维管束周围薄壁细胞核叶片薄壁细胞内。多糖在茎内分布最多,其次是叶片,根中分布较少。结论组织化学定位方法是一种研究和分析金钗石斛多糖的快速简便的技术和方法,金钗石斛叶片的开发多糖潜力较大,应充分利用。

Objective To optimize the methods of polysaccharide histochemistry localization for Dendrobimn nobile Lindl.and to determine it＇s ditribution and accumulation characteristic.Methods The cross Sctdion of root,stem and leaf of D.nobile were perifromed by means of freehand and frozen section method,the polysaccharide of D.nobile were analyzed by histochemistry localization.Results Freehand section was more appropriate for the study of histochemistry localization for pelysaccharide in D.nobile.The accumulation site of polysaccharide mainly distributed in the parenchyma cells of rot cortex,surrounding vascular bundle of stein,and in the parenchyma cells of leaf.The polysaccharide mainly distribuaed in stem,followed by leaf and root.Conclusion Histochemistry localization is a frogwent and simple mothod for analyzing the polysaccharide of D.nobile.The leaf of D.nobile plays suchan improtant role in the development of polysacharide that we should make full use of it.
