摘要
目的研究TGF-β1受体特异性拮抗剂SB-431542干预瘦素对肝星状细胞的生物活性及其相关机制。方法采用RT-PCR法检测α1(I)前胶原mRNA和α-平滑肌肌动蛋白(α-SMA)mRNA的变化;用间接免疫荧光来观察α1(I)前胶原蛋白、α-SMA蛋白的表达。结果瘦素加SB-431542组与瘦素组相比,各指标之间的差异有统计学意义(P<0.05),随浓度的增加,时间的延长,SB-431542对瘦素刺激的HSC-T6增殖纤维化有显著的抑制作用。结论 SB-431542可明显抑制瘦素的生物学活性,可能通过抑制TGF-β1信号系统而使细胞外基质(ECM)的合成减少,从而抑制肝纤维化。
【Objective】 Study on TGF-beta-1 receptor-specific antagonists SB-431542 intervention effect of leptin on biological activity of hepatic stellate cells and its mechanism.【Methods】 Detected by RT-PCR α1 collagen α1(ColIa1) mRNA and a-smooth muscle actin(α-SMA) mRNA changes;by indirect immunofluorescence to observe the α1(ColIa1) protein,α-SMA expression.【Results】 Leptin plus SB-431542 group compared with the leptin,the differences among the indexes were significantly(P 0.05),with concentration,time,SB-431542 on serum leptin stimulated the proliferation of HSC-T6 was significantly inhibited fibrosis.【Conclusion】SB-431542 can inhibit the biological activity of leptin,possibly by inhibiting the TGF-β1 signal system leaving the extracellular matrix(ECM) to reduce the synthesis of to inhibit liver fibrosis.
