摘要
目的研究冬凌草甲素(oridonin,ORI)对人食管癌细胞(ECCs)株Eca-109中食管癌干细胞的放射增敏作用。方法 32μg/mL冬凌草甲素处理的Eca-109细胞给予不同剂量照射,MTT法检测细胞增殖抑制作用,克隆形成实验检测细胞放射敏感性和克隆形成能力,流式细胞仪检测p75NTR的表达。结果实验组和对照组增殖抑制率组间比较差异有统计学意义(P<0.01)。克隆形成实验放射增敏比为2.35。随着照射剂量的增加,实验组和对照组p75NTR的表达均呈增加趋势;0、2和4 Gy剂量照射后实验组p75NTR的表达均少于对照组。结论 ORI对Eca-109细胞有明显的增殖抑制作用和放射增敏作用,其增敏机制可能与增加食管癌干细胞的放射敏感性有关。
【Objective】 To investigate the radiosensitization effect of oridonin on esophageal cancer stem cells of human esophageal cancer cell lines Eca-109.【Methods】 After irradiation with 0,2,4,6,8 Gy on the human esophageal cancer cells Eca-109 which cultured with oridonin(32 μg/mL),the proliferation inhibitory rate was measured by MTT assay.Meanwhile,the cell radiosensitivity and cloning formation ability were explored via clone formation assay.The expression levels of surface marker p75NTR of human esophageal cancer stem cell were also analyzed by flow cytometry.【Results】 The proliferation of esophageal cancer Eca109 cells was significantly inhibition by oridonin.The SF2,D0 and N values of the oridonin group were all lower than those of the control.Followed with the irradiation dose increase,the expression levels of surface marker p75NTR were increased.And oridonin groups compared with simple irradiation group in 0,2,4 Gy dose,the expression levels of p75NTR had a different decrease.【Conclusion】 Oridonin had significant proliferation inhibition and radiosensitization effect on esophageal cancer cell line Eca-109 cells,and this sensitization mechanism may be related to increased the radiosensitivity of esophageal cancer stem cells.
