摘要
通过苗期抗病基因推导,进一步利用FHPQ接种‘烟农15’和‘Thatcher’、‘ARCIN’和‘郑州5389’杂交得到的F2代群体进行抗叶锈鉴定,并进行标记筛选,明确‘烟农15’和‘ARCIN’中的抗叶锈病基因,并找到与之紧密连锁的分子标记。经苗期基因推导发现,这2个品种对多数叶锈菌小种表现为抗病,遗传分析结果表明,2个F2代群体的抗感分离比例均符合3∶1,表明这2个小麦品种的抗叶锈病基因由1对显性基因控制。利用标记分析结果表明,2个品种所含有的抗叶锈病基因与5DL染色体上与Lr1共分离STS的标记紧密连锁。结合基因推导、遗传分析和分子标记,表明这2个品种均携带抗叶锈病基因Lr1。
The objective of this study was to identify leaf rust resistance gene and its closely linked molecular markers in 'Yannong 15' and 'ARCIN'.After gene postulation in seeding,the two F1 and F2 populations from cross of Yannong15/Thatcher and ARCIN/Zhengzhou 5389,respectively,were inoculated with pathotypes FHPQ.The results indicated that two cultivars were resistant to most of P.triticina pathotypes,and the two F2 populations segregated with 3∶1,indicating the two wheat lines carried a single dominant resistance gene.By using the STS marker the leaf rust resistance gene in two lines is co-segregate with STS marker WR003,which was closely linked to Lr1.Based on the results from gene postulation,genetic analysis and molecular marker,it can be concluded that these two cultivars contain leaf rust resistance gene Lr1.
