血管瘤组织中p16基因甲基化的初步研究

摘要 目的探讨p16基因CpG岛甲基化在人血管瘤发生、发展及消退过程中的表达状况及其生物学意义。方法收集我科2008年至2010年手术切除血管瘤37例,其中男18例,女19例,年龄2个月~5岁。按Mulliken分类标准进行分组,增生期血管瘤21例,消退期血管瘤16例,另取瘤组织周围正常组织5例作对照。采用甲基化特异性PCR技术检测人血管瘤增生期、消退期及正常组织中p16基因CpG岛甲基化的表达频率。结果血管瘤标本的p16基因CpG岛甲基化率为40.5%(15/37),正常组织标本中未发现p16基因CpG岛甲基化,差异有统计学意义(P〈0.05)。增生期血管瘤组织中p16基因甲基化频率47.6%(10/21)高于消退期血管瘤组织中p16基因甲基化频率31.3%(5/16),差异有统计学意义(P〈0.05)。结论p16基因CpG岛甲基化可能在血管瘤发生、发展及消退过程中起着重要作用。 Objective To study the CpG islands methylation status of p16 gene in human he- mangioma. Methods From 2008 to 2010, 37 resected hemangioma tissues collected from 18 male pa- tients and 19 female patients were recruited in this study. The 19 patients aged from 2 months to 5 years old. According to the Mulliken classification, there were 21 proliferating hemangiomas and 16 involuting hemangiomas. Five tissue harvested form the normal tissues surrounding the hemangiomas were taken as normal controls. Methylation-specific PCR (MSP) was used to examine the detailed se- quence specificity of DNA methylation in the CpG island promoter/exon 1 region in the p16 gene in DNA from the hemangiomas. Results The CpG islands methylation of p16 gene was found on 15 out of the 37 hemangiomas patients(40. 5%). However, the CpG islands methylation of p16 gene was not found in any normal control tissues. The CpG islands methylation of p16 gene was found on 10 pa- tients with proliferating hemangiomas, which was significantly higher than that of the involuting he- mangomas(47. 6% vs. 31.3 %,P〈0. 05). Conclusions These data suggest that DNA methylation of the p16 gene in hemangioma could be the result of an active process that generates a discrete methyla- tion pattern and, hence, could be amenable to therapeutic manipulation.
