栀子对糖尿病大鼠肾脏的保护作用 被引量:4

摘要 目的探讨栀子水提取物、环烯醚萜总苷、京尼平苷、西红花苷对实验性糖尿病肾病(DN)大鼠的肾脏保护作用。方法采用高糖高脂饮食+链脲佐菌素腹腔注射,造成大鼠2型糖尿病模型。给药组分别灌胃相应药物,栀子水提取物组(2 g/kg)、环烯醚萜苷组(0.03 g/kg)、京尼平苷(0.003 g/kg),西红花总苷(0.02 g/kg)、洛汀新组(2 mg/kg)。连续给药4 w。检测:①血糖;②24 H尿微量白蛋白;③血清中尿酸、肌酐及尿素氮含量;④肾脏指数(肾重/体重比值);⑤光学显微镜下观察肾脏组织病理形态学变化,测量肾小球截面积(MGA)、细胞外基质截面积(MMA),计算MMA/MGA比值。结果给药4 w后,用药后与用药前比较,栀子水提物、栀子总苷组和西红花苷组血糖均有所差异(P<0.05)。用药后各给药组与模型组比较,血糖有所下降,但差异不明显。栀子各给药组24 h尿蛋白定量较模型组均有显著降低(P<0.01)。与模型组比较,给药组肾脏指数均有下降,其中京尼平苷组、西红花苷组肾脏指数下降较明显(P<0.05)。栀子水提物、京尼平苷和西红花苷均对大鼠血清尿酸水平有所降低(P<0.05),栀子水提物能显著降低大鼠尿素氮水平(P<0.01),西红花苷对大鼠尿素氮水平也有正性影响(P<0.05);栀子各给药组对大鼠血清肌酐水平影响不大。光镜示栀子各给药组肾脏组织病理损害较模型组有明显改善,栀子水提物组和京尼平苷组MMA/MGA值较模型组有所降低(P<0.05)。结论栀子各提取物对DN大鼠具有较为明显的肾脏保护作用。 Objective To observe the renal protection effect on rats of the diabetic nephropathy treated by water extract of gardenia, iridoid glycosides', geniposide and saffron total glycosides. Methods Setting up diabetic rats model by high-sugar and high-fat diet and in-traperitoneal injection of streptozotoein. Intragastric administration of the corresponding drugs were given for 4 weeks. Dose of different drugs were the water extract of Gardenia jasminoides (2 g/kg), gardenia total glycosides (0. 03 g/kg), genipin (0. 003 g/kg), crocin (0. 02 g/ kg), lotensin (2 mg/kg) respectively. Blood glucose, urinary protein, uric acid (UA), creatinine (Cr), blood urea nitrogen (BUN), kid-ney coefficient calculation were detected. Pathological Changes of kidney was observed and the MGA, MMA were recorded. Results After 4 weeks of administration, compared with before treatment, blood glucose of three group treated by the water extract of Gardenia jasminoides, Gardenia total glycosides and erocin were decreased ( P 〈 0. 05 ). Compared with model group after treatment, the change was little. The 24 h urinary protein of each group treated by Gardenia was lower than that of model group ( P 〈 0.01 ). The kidney coefficient calculation (K/W) also was decreased, the change of K/W in genipin and croein group were apparent (P 〈0. 05). Serum UA in the water extract of Gardenia, genipin and croein groups were significantly decreased ( P 〈 0. 05 ) , BUN were decreased by the water extract of Gardenia ( P 〈 0. 01 ) and croein ( P 〈 0. 05 ). Gardenia had little effect on Ser. Pathological changes of kidney in each group treated by Gardenia were less than those of model group. MMA/MGA also was decreased, the change of water extract of Gardenia and genipin group were apparent (P 〈 0.05 ). Conclusions Gardenia extract has some positive effect on diabetic mice's blood sugar and kidney function.
