摘要
目的探讨单核细胞趋化蛋白-1(MCP-1)和骨形成蛋白-7(BMP-7)在耳部瘢痕疙瘩发病机制中的作用和意义。方法采用免疫组织化学方法检测MCP-1和BMP-7在39例耳部瘢痕疙瘩、20例耳部非病理性瘢痕和20例正常耳部皮肤组织中的表达水平,并对耳部瘢痕疙瘩组织中MCP-1和BMP-7的表达情况进行相关性分析。结果与耳部非病理性瘢痕和正常皮肤组织相比,MCP-1在耳部瘢痕疙瘩组织中表达呈显著性上调(P<0.01),而BMP-7的表达呈显著性下调(P<0.01),两者呈显著性负相关(r=-0.54,P<0.01)。结论 MCP-1的过度表达和BMP-7的表达下调共同参与了耳部瘢痕疙瘩的纤维化病理过程;BMP-7是一个极有前途的基因治疗靶点。
Objective To investigate the expression of monocyte chemoattractant protein-1 (MCP-1) and bone mor- phogenetic protein-7 (BMP-7) in auricular keloids and their roles in the pathogenesis of auricular keloids. Methods The ex- pression of MCP-1 and MBP-7 was detected using immunohistochemistry methods in auricular skin tissues from 20 normal controls, in non-keloid scar tissues from 20 patients after middle ear surgeries and in auricular keloid from 39 patients. Cor- relation of expression among different tissues was analyzed. Results A significantly increased expression of MCP-1 was found in auricular keloids (P 〈0.01),while the expression of MBP-7 was the lowest in keloids (P 〈0.01), in comparison with non-keloid auricular scars as well as normal auricular skins. MCP-1 and MBP-7 expression was negatively correlated (r=-0.54,P〈0.01). Conclusion The up-regulated expression of MCP-1 and the down-regulated expression of MBP-7 may together promote the pathological process of fibrosis in auricular keloids. MBP-7 might be a promising gene therapy target.
