摘要
目的研究抗淋巴细胞球蛋白(Antilymphocyte Globuli ALG)经内耳给药后能否改善豚鼠内耳免疫反应动物模型的免疫损伤。方法 24只清洁级健康Hartley豚鼠随机分为三组,即试验组、阳性对照组、阴性对照组(n=8),试验组、阳性对照组以钥孔戚血蓝素(keyhole limpet hemocyanin KLH)建立右耳免疫反应动物模型,阴性对照组右耳以PBS对照;建模后第3天试验组、阴性对照组予内耳注射兔抗鼠免疫球蛋白,阳性对照组予PBS对照。通过观察听性脑干反应(auditory brainstem response ABR)、外淋巴液抗KLH抗体,了解ALG对内耳异常免疫调节作用。结果造模后3天试验组及阳性对照组听力较阴性对照组有明显下降(p﹤0.05),ALG干预后1周,试验组听力较阳性对照组有明显改善(p﹤0.05),4周后与阴性对照组无明显差异(p﹥0.05);在KLH免疫后外淋巴抗KLH抗体升高,ALG可阻止或减轻抗KLH抗体在内耳的表达(p﹤0.05)。结论 ALG可阻止或减轻内耳异常免疫反应,对梅尼埃病等内耳病症有临床应用前景。
Objectlve To study the effects of Antilymphocyte Globulin (ALG) on inner ear immune injury in guinea pigs. Methods Twenty four clean grade health Hartley guinea pigs were randomly and evenly divided into a treatment group, a posi- tive control group and a negative control group. Right ear immune reaction was induced with keyhole limpet hemocyanin (KLH) in animals in the treatment and positive control groups, while those in the negative control group were treated with PBS. Three days after modeling, rabbit anti mouse immune globulin were injected into the inner ear in animals in the treatment and negative control groups, while PBS was injected in animals in the positive control group. ALG effects on immune function regu- lation were determined through measuring auditory brainstem responses (ABRs) and perilymphatic anti KLH antibodies. Results There was significant hearing loss in the treatment and positive control group after modeling, as compared with the negative control group, (p 〈 0.05). One week after ALG treatment, hearing was significantly improved in the treatment group, as com- pared with the positive control group (p 〈 0.05). and was similar to that in the negative control group 4 weeks later (p 〉 0.05)., Perilymphatic anti-KLH antibody levels increased after KLH immunization, indicating ALG' s protective effects against ex- pression of anti-KLH antibody in the inner ear (p 〈 0.05).Conclusion ALG appears to prevent or mitigate abnormal inner ear immune responses. It may have clinical application prospects for inner ear disease such as Meniere's disease.
