摘要
目的研究塞来昔布对人鼻咽癌细胞增殖的抑制作用。方法以人鼻咽癌细胞株CNE为研究对象,MTT法观察不同浓度和作用时间的塞来昔布对细胞增殖的抑制作用;用CNE细胞株建立裸鼠移植瘤模型,通过测量瘤体体积及重量,计算对移植瘤生长的抑制效果。结果 MTT结果显示塞来昔布对CNE细胞株生长有显著的抑制作用且呈现时间和剂量的依赖性,在体动物实验表明,实验组与对照组相比瘤体的体积与重量差异有统计学意义。结论环氧合酶(cyclooxygenase-2,Cox-2)抑制剂塞来昔布对人鼻咽癌细胞有明显增殖抑制作用,可能是预防和治疗鼻咽癌的重要候选药物之一。
OBJECTIVE To study the inhibitory effects of celecoxib on the proliferation of nasopharyngeal carcinoma cells. METHODS The inhibitory effects of celecoxib at different concentrations and different treatment time on nasopharyngeal carcinoma cell line CNE were observed with MTT assay. The inhibition effect of celecoxib was evaluated in mice bearing transplanted tumor derived from nasopharyngeal carcinoma cell line CNE transplantation in vivo. RESULTS Celecoxib significantly inhibited the growth of nasopharyngeal carcinoma cell line CNE in a time-and dose-dependent manner. In the mice bearing CNE transplanted tumor, celecoxib showed significant inhibitory effect on the growth of the transplanted tumor. CONCLUSION Celecoxib can inhibit the proliferation of different nasopharyngeal carcinoma cell lines both in vitro and in vivo, and therefore it may serve as an important candidate drug for prevention and treatment of nasopharyngeal carcinoma.
