Synthesis of artificial methadone antigen

目的:利用化学方法将美沙酮半抗原与载体蛋白偶联制备美沙酮人工抗原。方法:结构修饰后的美沙酮类似物与载体BSA蛋白经过混合酸酐法或碳二亚胺法连接合成美沙酮人工抗原,利用紫外光谱扫描法、蛋白质电泳法和胶体金免疫层析法进行检测。结果:美沙酮人工合成抗原连接成功,胶体金免疫层析检测结果显示合成抗原与抗体有特异性反应。结论:该方法合成的美沙酮抗原可作为美沙酮免疫用抗原和美沙酮免疫检测方法建立所用包被抗原。

Objective：To synthesize Methadone-bovine serum albumin by chemical process.Methods：Methadone＇s analog synthesized by us was conjugated to protein by mixed anhydride or EDC method,and the carrier protein was BSA.The result was confirmed by UV scanning method,protein electrophoresis and gold immunochromatography assay（GICA） methods.Resulsts：The conjunction was successful and the formed conjugate showed confirmed antigenic activity.Conclusion：Methadone antigen can be synthesized successfully by above method,which can be used as immunity antigen and coated antigen in immunoassay for Methadone.
