摘要
目的 研究中药黄芪对外周血造血干细胞移植术后早期造血功能重建有无促进作用。方法 以致死量辐照并行外周血造血干细胞移植的 BAL B/ c小鼠为模型 ,移植后给予黄芪注射液 10 g/ kg或 2 0 g/ kg腹腔注射 ,或粒细胞 -集落刺激因子 ( G- CSF,商品名 :惠尔血 ) 2 0 0μg/ kg皮下注射 ,连续 15天 ,观察 4周内外周血常规的恢复情况 ,并对骨髓造血微环境行透射电镜观察。结果 黄芪组、惠尔血组小鼠外周血白细胞的恢复均快于对照组 ,但黄芪组慢于惠尔血组 ,血小板的恢复各组间无显著差异 ,两个黄芪剂量组间无显著差异 ;电镜检查发现黄芪组小鼠骨髓内皮细胞、网状细胞的损伤轻于对照组和惠尔血组 ,内皮细胞、网状细胞与造血原始细胞间的接触也较对照组和惠尔血组紧密。结论 黄芪能促进小鼠外周血造血干细胞移植术后早期白细胞的重建 ,其机理可能是通过保护和改善骨髓造血微环境 ,改善骨髓基质细胞与造血干细胞间的接触 ,并促进 G- CSF等内源性细胞因子的分泌 ,从而促进干细胞的增殖 ,而惠尔血则直接作用于干细胞 。
Objective To study the effects of Astragalus, a Chinese herbal medicine, on short term hemopoietic reconstitution after peripheral blood stem cell transplantation.Methods A model was formed with BALB/c mice, which received mobilized peripheral blood stem cells from donor BALB/c mice after irradiation with a lethal dose. The recipients were treated with Astragalus 10g/kg or 20g/kg i.p.or Filgrastim 200μg/kg s.c. for 15 days after transplantation. The recovery of the peripheral blood cells was monitored in four weeks. The bone marrow specimens were collected on day 15, and observed under transmission electron microscope (TEM).Results Both the Astragalus groups and the Filgrastim group had a more rapid recovery on white blood cell than the control group, but the recovery was slower in the Astragalus groups than in the Filgrastim group. It was similar on the recovery of platelet in all the groups, and there was no difference on hemopoietic reconstitution between the two Astragalus groups. The results of TEM showed that the bone marrow endotheliocytes and reticulocytes had less impairment in the Astragalus group than in the control group and Filgrastim group, and had a better contact with the hemopoietic original cells.Conclusion This study suggested that Astragalus may accelerate the short term reconstitution of white blood cell in mice after proipheral blood stem cell transplantation, protect and improve the bone marrow hemopoietic microenvironment, improve the contact between the stromal cells and the stem cells, facilitate the secretion of endogenous hemopoietic growth factors, and consequently accelerate the expansion of the stem cells, which is different from the direct effect of Filgrastim on the expansion of the stem cells.
