摘要
目的 应用 Kaneko兔异位心肺移植模型研究苯肾上腺素预处理在兔心肺保存中的作用。方法 32只健康雄性青紫蓝兔随机分为两组 ,各 16只。实验组心肺获取前给予苯肾上腺素 5 0μg· kg- 1 预处理 ,对照组给予等量生理盐水 ,两组心肺均保存 4h后移植。心肺移植体转为自主功能状态后 ,连续观察 2 h,每 30 min测定左室收缩压、左室舒张末压、右房压、动脉血氧分压 ,以评定移植体功能。实验结束时取心肺组织测定丙二醛 ( MDA)和超氧化物歧化酶 ( SOD)的含量。结果 实验组心肺功能优于对照组 ,心肺组织 MDA含量低于对照组 ,SOD含量高于对照组。结论 苯肾上腺素预处理可加强低温保存时兔心肺保护作用 。
Objective Using Kaneko′s rabbit heartlung transplantation model, to investigate the effects of preservation of heartlung graft in rabbit by precondition with intravenous phenylephrine. Methods Thirtytwo healthy male rabbits were randomly divided into experimental group and control groups (16 rabbits in each group). The donors of the experimental group were intravenously administrated phenylephrine (50 μg·kg -1 ) before the harvest of grafts. The donors of control group received normal saline. After heartlung transplantation, the donors′ grafts LVSP, LVedp, RAP, PaO 2 were measured every 30 minutes to evaluate the function of heart and lung, The contents of MDA and SOD of grafts′ tissue were measured finally. Results The of heart and lung has better in experimental group than in control group (P<0.05). The experimental group had less MDA and higher SOD contents than the control group. Conclusion The phenylephrine preconditioning in combination with cold preservation was superior to cold preservation alone on preservation of heartlung grafts. The possible mechanism is related to reduction of production of oxygen free radicals.
