摘要
目的:研究绞股蓝总皂苷含药血清对皮肤光老化HaCaT细胞分泌炎症因子IL-1β、IL-6表达的影响。方法:选取20只大鼠,随机分为四组,每组5只,分别设为空白血清组、低剂量绞股蓝总皂苷含药血清组、中剂量绞股蓝总皂苷含药血清组、高剂量绞股蓝总皂苷含药血清组,采用UVB照射人工模拟光老化HaCaT细胞模型,照射后用含不同浓度绞股蓝总皂苷的大鼠血清进行干预,12 h后采用ELISA方法检测各组细胞分泌IL-1β、IL-6的表达水平。结果:与空白对照组比较,UVB模型组细胞IL-1β、IL-6表达显著上调;与UVB模型组比较,低剂量绞股蓝总皂苷含药血清组、中剂量绞股蓝总皂苷含药血清组、高剂量绞股蓝总皂苷含药血清组细胞中IL-1β、IL-6表达显著下调(P<0.01)。结论:绞股蓝总皂苷含药血清可能通过抑制UVB辐射人皮肤角质细胞诱导炎症信号分子防护皮肤光老化。
Objective: To study the effect of gypenosides-containing serum on secretion of inflammatory factors IL-1 β,IL-6 of photoaging skin HaCaT cells.Methods: 20 rats were randomly divided into four groups with 5 rats in each group: blank serum group,low dose of gypenosides-containing serum group,middle dose of gypenosides-containing serum group,high dose of gypenosides-containing serum group.After UVB irradiation,the artificial light aging of HaCaT cell models were interfered with rat serum containing different concentrations of Gypenosides,12 h later the expression level of cell secretion of IL-1 β,IL-6 were determined with ELISA.Results: Compared with the blank control group,IL-1β,IL-6 expression of UVB model group was significantly higher;compared with UVB model group,IL-1 β,IL-6 expression in cells of low dose group,middle dose group,high dose group was significantly lower(P0.01).Conclusion: Gypenosides-containing serum can protect skin photoaging by inhibiting the UVB radiation of human skin keratinocytes.
