摘要 目的研究苹果多酚、蓝莓花色苷和天山雪莲总黄酮三种不同来源的植物化学物对巨噬细胞白血病细胞系RAW264.7的生长抑制作用及比较三种不同来源的类黄酮和多酚类植物化学物对细胞因子的影响。方法噻唑蓝(MTT)比色法检测不同作用剂量的三种物质对RAW264.7细胞的影响;逆转录聚合酶链反应(RT-PCR)方法检测三种物质处理不同时间(24 h、48 h)对肿瘤坏死因子α(TNF-α)、白介素6(IL-6)、环氧化酶2(COX-2)等细胞因子mRNA表达的影响;免疫印迹法检测三种物质对ERK1、2蛋白的影响。结果三种物质均能诱导TNF-αmRNA的表达,表达量与作用时间呈现正相关;天山雪莲总黄酮对IL-6 mRNA诱导作用最明显,三种物质在48 h作用点抑制COX-2 mR-NA表达水平作用明显强于24 h。同时天山雪莲总黄酮能减少RAW264.7细胞中ERK1、2蛋白表达量。结论天山雪莲总黄酮对RAW264.7细胞影响最为明显,而蓝莓花色苷和苹果多酚的影响程度弱于天山雪莲总黄酮。 Objective To study the effect of anthocyanin, apple polyphenol and Sanssurea Medusa Maxim on cell proliferation and eyto- kines in RAW264.7 cell line. Methods The influence of three chemicals on RAW264.7 cell line, TNF - α, IL - 6 , COX - 2 eytokine and ERK1, 2 protein were examined by MTF, RT - PCR and western blot at 24 and 48 h of treatment. Results These results showed that three chemicals could induce the expression of TNF - α mRNA and there was positive correlation between expression density and treatment time. It was apparent that flavonoids of Tianshan snow lotus could induce the expression of TNF - α mRNA, anthoeyanin, apple polyphenol and Saussurea Medusa Maxim showed growth inhibition effect on RAW264.7 cells and decreased expression level of COX - 2 mRNA and decreased amount of ERK1, 2 protein in RAW264.7 cells. Conclusion The effect of Tianshan snow lotus on RAW264.7 cell line is most significant, and the effect of blueberry anthocyanins and apple polyphenols is weaker than that of total flavonoids of Tianshan snow lotus.
