干燥综合征患者外周血淋巴细胞Toll样受体4CD14的表达 被引量:2

The expressions of TLR4 and CD14 in peripheral blood monouclear cells in patients with Sjogren's syndrome

摘要 目的探讨干燥综合征(SS)患者外周血淋巴细胞Toll样受体(TLR)4、CD14的表达。方法分别采用反转录-聚合酶链反应(RT-PCR)法和流式细胞仪检测TLR4、CD14mRNA和蛋白的表达。结果 SS患者外周血单个核细胞TLR4、CD14mRNA和蛋白的表达均高于对照组(P<0.05);初发组TLR4、CD14mRNA和蛋白表达水平明显高于缓解组(P<0.05);复发组TLR4、CD14mRNA和蛋白表达水平与缓解组相比明显增高(P<0.05);复发组TLR4、CD14mRNA和蛋白表达水平与初发组相比无统计学意义(P>0.05)。结论①SS患者外周血淋巴细胞高表达TLR4和CD14,且与病情活动相一致;②脂多糖(LPS)-TLR4信号传导途径参与了SS的发病机制。 Objective To investigate the expressions of TLR4 and CD14 in peripheral blood monouclear cells(PBMCs) in patients with Sjogren's syndrome(SS).Methods The expressions of mRNA and protein of TLR4 vs CD14 were detected by reverse transcription reaction(RT-PCR) and flow cytometry(FCM) respectively.Results The expressions of mRNA and protein of TLR4 vs CD14 in patients with SS were significantly higher than those in controls(P〈0.05);The expressions of mRNA and protein of TLR4 vs CD14 in initial and recurrence groups were significantly higher than those in remission group(P〈0.05);There was no difference in expressions of mRNA and protein of TLR4 vs CD14 between recurrence group and initial group(P〈0.05).Conclusion ①The PBMC patients with SS had the highly expressions of TLR4 and CD14 in blood lymphocyte,which was consistent with disease activity.② LPS-TLR4 signal pathway have participated into pathogenesis of SS.
