摘要
质粒pMC71A-LEU在对阴沟肠杆菌E26-1(EnterobactercloacaeE26-1)的转化中研究了感受态细胞、CaCl2浓度、热激温度和质粒DNA量对转化频率的影响。结果表明,在阴沟肠杆菌F26-1生长到OD600=0.45时,用50mmol/LCaCl2处理细胞,加入100ng质粒DNA,在32℃热激2min,转化频率最高,可达1×103转化子/μgDNA。
The effect of plasmid pMC71A-LEU to receptive cells, concentrtion of CaCl2, heat stimulated temperature and the arnotint of plasmid DNA in transformation frequency of Enterobacter cloacae E26-1 was studied. The results indicated that when E. chacae grew at OD600 = 0. 45, the transformation frequency was the highest, as high as 1 × 10 3 transformants/μg,when the cells treaed with 50 mmol/L of CaCl2, adAn with 100ng of plasmid DNA, and heat stimulation at 32 ℃ for 2 minutes.
