工业化方法提取及纯化豆渣RNA的工艺研究 被引量:1

Study on the Industrial Methods of Extraction and Purification RNA from Bean Dregs

摘要 以新鲜豆渣为原料,在单因素试验基础上,根据中心组合试验设计原理进行四因素三水平的响应面分析试验,对豆渣中提取RNA的工艺参数进行优化,同时对RNA的最优沉淀剂进行了筛选。结果表明,其最适裂解条件为裂解温度92℃,裂解时间70 min、裂解液pH 5.4、液料比10∶1,且在此条件下的最适沉淀剂为异丙醇-高NaCl溶液。此法最终的豆渣RNA得率为0.48%,OD260/OD280为1.83。 Bean dregs were used as materials. Based on the results from the university test, central composite design combined with response surface test were employed to optimized parameters of extraction process, and selected optimal RNA precipitation agent. The results showed that optimal extraction of RNA was: extraction temperature at 92 ℃, water bath extraction 70 rain, pH at 5.4, and fixed substrate concentration at 1 : 10. In these conditions the optimums for precipitation agent were isopropanol-saturated solution of NaC1. Under the above conditions, the yield of RNA was up to 0.48%, 00260/ D280 was 1.83. The study laid the foundation for the industrial production and soybean resources comprehensive utilization.
