摘要
目的研究不同年龄组大鼠心脏血管紧张素Ⅱ1型受体(AT1R)和血管紧张素Ⅱ2型受体(AT2R)对α1肾上腺素受体(α1-AR)信号转导的影响。方法选择Wistar大鼠29只,根据不同月龄,分为3.5月龄组8只、12月龄组8只、18月龄组7只和24月龄组6只。检测大鼠心肌组织α1-AR与AT1R和α1-AR与AT2R蛋白激酶C活性;并分别与α1-AR单独激动时蛋白激酶C活性进行比较。结果与激动α1-AR后比较,激动AT1R和α1-AR后,各月龄组大鼠心肌α1-AR介导的蛋白激酶C活性均明显升高,差异有统计学意义(P<0.05,P<0.01);与激动α1-AR后比较,激动AT2R和α1-AR后,3.5月龄组、12月龄组大鼠心肌α1-AR介导的蛋白激酶C活性差异无统计学意义(P>0.05),18月龄组和24月龄组大鼠心肌α1-AR介导的蛋白激酶C活性明显降低,差异有统计学意义(P<0.05,P<0.01)。结论 AT1R在各年龄组均使α1-AR的信号转导途径有增强作用;在心脏衰老过程中,AT2R对α1-AR的信号转导途径有抑制作用。
Objective To study the effect of types 1 and 2 angiotensin II receptor(AT1R,AT2R) on a1 adrenoceptor(a1-AR) signal transduction in heart of rats at different ages. Methods Twentynine Wistar rats were divided into 3.5 months old group(n=8),12 months old group(n=8),18 months old group(n=7) and 24 months old group(n= 6). Protein kinase C activity in a1-AR, AT1 R and AT2 R agitated rat myocardial tissues was measured and compared with that in a1-AR agitated rat myocardial tissues. Results The protein kinase C activity was significantly higher in different months old groups after a1-AR agitation than after AT1R and a1-AR agitation, and sig nificantly lower in 18 months old group and 24 months old group after AT2R and a1-AR agitation than before AT2R agitation(P〈0.05,P〈0.01). No significant difference was found in a1-AR mediated protein kinase C activity between 3.5 months old group and 12 months old group(P〉 0.05). Conclusion AT1R can increase the a1-AR signal transduction in rats at different ages and inhibit it in aging heart of rats.
