摘要
甜瓜(Cucumis melo)子叶外植体能被含双元载体的根瘤农杆菌菌株LBA4404感染。该载体内含1个NPT-Ⅱ基因(新霉素磷酸基转移酶Ⅱ基因),以供选择卡那霉素抗性的转化甜瓜细胞和1个WMV-2 CP基因(西瓜花叶病毒2号外壳蛋白基因)。卡那霉素抗性植株经PCR-Southem杂交试验表明,外源的WMV2CP基因确已通过农杆菌介导的转化途径导入甜瓜细胞。目前转基因甜瓜已开花结果。攻毒试验表明,与对照相比,转基因植株可以推迟发病时间,减轻发病程度,表现出对WMV-2具有较强的抗性。
Segments of cotyledon explants was infected by Agrobacterium tumefaciens harboring a binary vector that contains a NPT-II gene and a WMV-2 CP gene. Transgenic plants were obtained. Successful genetic transformation was confirmed by PCR-Southem blot. The results of the resistance test of transgenic Cucumis melo to WMV-2 show that transgenic plants can delay the disease infection and reduce the incidence of virus diseases, thus showing certain resistance to relevant virus.
