生防木霉SS003菌株(Trichoderma atroviride)的固体发酵工艺研究

The Solid Fermentation Technology for Bio-control Strain SS003 of Trichoderma atroviride

为了探索酷绿木霉SS003菌株固体发酵产孢子的较优工艺条件。分别以酷绿木霉SS003菌株为供试菌和以玉米秸秆、麦麸发酵基质,分别进行了固体发酵培养基配方和固体发酵条件的优化筛选,试验设计均采用单因索试验。试验最终检测方法采用血球计数板倍量稀释法镜检孢子量(108个/g)。分别获得了酷绿木霉SS003菌株产孢固体发酵的较优培养基配方和较优固体发酵条件,即80目的玉米秸秆与麦麸配比1∶3,接种1×106个/mL的SS003孢子液,保持含水量55%和充分通气,培养11 d后,酷绿木霉SS003菌株固体发酵产孢量达到最大(76.14×108个/g)。初步探索到绿木霉SS003菌株固体发酵产孢的较优工艺条件,为该菌的深入研发提供了有益的试验数据。

To explore the better technology for the spore production of the strain SS003 of Trichoderma atroviride.Strain SS003 of Trichoderma atroviride as testing strain and corn straw and wheat bran as substrates,the methods of single-factor selection were adopted to optimize and select the substrate ingredients and conditions for the solid fermentation.The final test adopted the hemocytometer doubling-dose dilution method to microscopically examine numbers of bacterial colonis（108/g）.Comparatively good substrate ingredients for the spore production of the strain SS003 of Trichoderma atroviride and good conditions for solid fermentation were as follows：to inoculate 1×106个/mL spore suspension of the strain SS003 to the substrates with the ratio of corn straw and wheat bran through 80 mesh sieve was 1∶ 3,maintaining content of moisture as 55% and good ventilation,after 11 d,the spore production of the strain SS003of Trichoderma atroviride under solid fermentation reached maximum at 76.14×108/g.The comparatively good technology for the spore of the strain of Trichoderma atroviride under solid fermentation was primarily obtained,which would benefit its deep research.
