摘要
目的探讨Akt和mTOR蛋白是否可作为一种新的诊断肾透明细胞癌标志和药物靶向治疗的靶点。方法采用免疫组化和逆转录聚合酶链反应(RT-PCR)技术对60例肾透明细胞癌组织和30例癌旁正常肾组织中的Akt和mTOR的蛋白及mRNA表达进行检测分析。结果 Akt和mTOR蛋白及mRNA在肾透明细胞癌组织中均呈高表达,在癌旁正常组织中均呈低表达(P<0.05);mTOR和Akt蛋白在肾细胞癌组织中的阳性表达率呈正相关。结论 Akt、mTOR蛋白在肾透明细胞癌中的高表达与肾细胞癌的发生有关,可作为一种新的肾细胞癌诊断标志物。
Objective To explore an applicable marker for diagnosis and a target to develop therapeutic agents for renal clear cell carcinoma. Methods mTOR, Akt protein and mRNA expression in carcinoma tissues in 60 patients and adjacent tissues in 30 patientswith clear cell renal cell carcinoma were detected by immunohistochemistry (IHC) and reverse transcriptase polymerase chain reaction ( RT - PCR ). Results The results showed the positive expression rate of Akt in carcinoma tissue was significantly higher than adjacent tissues; the positive expression rate of mTOR in carcinoma tissue was significantly higher than that in adjacent tissues (P 〈 0.05 ). RT - PCR results : mRNA expression of Akt and mTOR in carcinoma tissue was higher than that in adjacent tissues ; Most adjacent tissues were Positive for mRNA expression of Akt and roTOR in adjacent tissues. Conclusion mTOR and Akt protein expression may he an applicable marker for diagnosis and/or a target to develop therapeutic agents for clear cell renal cell carcinoma.
