利用种子储藏蛋白电泳分析小麦材料SY95-71及其亲本的遗传变异 被引量:5

摘要 运用种子储藏蛋白电泳方法 ,对来源于六倍体小黑麦Eronga83和普通小麦品种凡 6杂交培育的小麦材料SY95 - 71进行了遗传变异分析。酸性聚丙烯酰胺凝胶电泳 (APAGE)对种子醇溶蛋白的组成分析认为 ,与小麦亲本相比 ,SY95 - 71缺少了 1D染色体上的Gli-D1带 ,这在普通小麦品种 (系 )中较为少见 ,其他醇溶蛋白带均来自双亲 ,而与小黑麦亲本更为接近。SDS -聚丙烯酰胺凝胶电泳 (SDS -PAGE)对高分子谷蛋白亚基的组成分析认为 ,SY95 - 71的位点Glu -A1和Glu -D1与两个亲本不同 ,而Glu -B1来自Eronga83,这些遗传变异是来源于培育过程中的基因重组。另外对SY95 - 71的感条锈病性与储藏蛋白位点的变异的关系以及六倍体小黑麦对小麦种质创新的价值进行了讨论。 The Genetic variations of wheat line SY95-71 and its Parents, a hexaploid Triticale Eronga83 and wheat line Fan6, were investigated by using seed storage protein electrophoresis, The gliadin pattern by APAGE analysis shows that the Gli-D1 loci is lost in SY95-71, which is rare in common wheat. The most of gliadin loci of SY95-71 are from its patents but more similar to Eronga83. The HMW-GS loci by SDS-PAGE revealed that the Glu-A1 and Glu-D1 are different from in their parents, but the Glu-B1 is from Eronga83. The results indicated that the most variation in SY95-71 in comparison with their parents result from the gene recombination. In addition, the collation between the susceptibility to stripe rust and the gliadin variation in SY95-71 and the value of hexaploid Triticale to wheat germplasm are also discussed in this paper.
